Progress and challenges in genome-wide studies to understand the genetics of diabetic retinopathy

:-

Abstract: There are many advantages to understanding the genetics of human disease. Genetic markers can be used to calculate the risk of developing a disease, and elucidation of genetic risk factors can pinpoint the molecular aetiology of disease, which can facilitate the development of targeted therapies. Diabetic retinopathy (DR) is a common complication of diabetes that has a significant impact on quality of life. It has a clear genetic component, but determination of the genetic risk factors has proven difficult. To date, genome-wide studies for DR have been conducted on relatively small patient cohorts compared to other complex eye diseases and replication of genetic findings has been limited. The disease is highly heterogeneous, confounding attempts to classify patients into appropriate groups for genetic analysis and making direct comparisons between studies challenging. Future studies to determine the genetic causes of DR will need to focus on larger sample sizes, detailed phenotyping and appropriate classification of patients. Global co-operation and meta-analyses combining data from multiple studies will be critical to the discovery of genetic risk loci for DR.

An update of the Guangzhou Twin Eye Study

:-

Abstract: The Guangzhou Twin Eye Study (GTES) is a population-based study of young twins residing in Guangzhou City. The major aim of GTES is to explore the impact of genes, environmental factors and gene-environment interactions on common eye diseases. From 2006, for more than 1,300 twin pairs, age 7–26 years old, progressive ocular phenotypes, such as refraction, ocular biometrics, weight, and height were collected annually, while non-progressive phenotypes such as parental refraction, corneal thickness, retinal fundus, intraocular pressure and DNA only collected at baseline. In the current study, we summarize the major findings on the etiology of myopia in recent decades.

Strategies to control myopia in children: a review of the findings from the Anyang Childhood Eye Study

:-

Abstract: Myopia in children remains a major public health problem worldwide, especially in some Asian countries such as China, Singapore and Japan. Although many interventions have been attempted, few has been proven to be effective in controlling onset and progression of myopia in children. Environmental factors, genetic susceptibility or ethnic differences can affect the efficacy of these interventions. However, many questions remain unclear and even controversial for controlling myopia. China has the biggest population with myopia, especially for children myopia. Thus, it is of importance to present what achievements Chinese scientists have made in the field of myopia control in children. We summarize the current findings on myopia control in children from the Anyang Childhood Eye Study, including epidemiological data, clinical trials, systematic reviews and meta-analyses, and compare them with studies in other countries to find potential clues for controlling myopia in children.

Objective electrophysiological contrast sensitivity with monofocal and multifocal intraocular lenses: a prospective clinical study

:-

Background: To compare objective electrophysiological contrast sensitivity function (CSF) in patients implanted with either multifocal intraocular lenses (MIOLs) or monofocal intraocular lenses (IOLs) by pattern reversal visual evoked potentials (prVEP) measurements.

Methods: Fourty-five cataract patients were randomly allocated to receive bilaterally: apodized diffractive-refractive Alcon Acrysof MIOL (A), full diffractive AMO Tecnis MIOL (B) or monofocal Alcon Acrysof IOL (C). Primary outcomes: 1-year differences in objective binocular CSF measured by prVEP with sinusoid grating stimuli of 6 decreasing contrast levels at 6 spatial frequencies. Secondary outcomes: psychophysical CSF measured with VCTS-6500, photopic uncorrected distance (UDVA), and mesopic and photopic uncorrected near and intermediate visual acuities (UNVA and UIVA respectively).

Results: Electrophysiological CSF curve had an inverted U-shaped morphology in all groups, with a biphasic pattern in Group B. Group A showed a lower CSF than group B at 4 and 8 cpd, and a lower value than group C at 8 cpd. Psychophysical CSF in group A exhibited a lower value at 12 cpd than group B. Mean photopic and mesopic UNVA and UIVA were worse in monofocal group compared to the multifocal groups. Mesopic UNVA and UIVA were better in group B.

Conclusions: Electrophysiological CSF behaves differently depending on the types of multifocal or monofocal IOLs. This may be related to the visual acuity under certain conditions or to IOL characteristics. This objective method might be a potential new tool to investigate on MIOL differences and on subjective device-related quality of vision.

AB057. Diagnostic information for the recognition of 3D forms in humans

:-

Background: The perception of visual forms is crucial for effective interactions with our environment and for the recognition of visual objects. Thus, to determine the codes underlying this function is a fundamental theoretical objective in the study of the visual forms perception. The vast majority of research in the field is based on a hypothetico-deductive approach. Thus, we first begin by formulating a theory, then we make predictions and finally we conduct experimental tests. After decades of application of this approach, the field remains far from having a consensus as to the traits underlying the representation of visual form. Our goal is to determine, without theoretical a priori or any bias whatsoever, the information underlying the discrimination and recognition of 3D visual forms in normal human adults.

Methods: To this end, the adaptive bubble technique developed by Wang et al. [2011] is applied on six 3D synthetic objects under varying views from one test to another. This technique is based on the presentation of stimuli that are partially revealed through Gaussian windows, the location of which is random and the number of which is established in such a way as to maintain an established performance criterion. Gradually, the experimental program uses participants’ performance to determine the stimulus regions that participants use to recognize objects. The synthetic objects used in this study are unfamiliar and were generated from a program produced at C. Edward Connor’s lab, Johns Hopkins University School of Medicine.

Results: The results were integrated across participants to establish regions of presented stimuli that determine the observers’ ability to recognize them—i.e., diagnostic attributes. The results will be reported in graphical form with a Z scores mapping that will be superimposed on silhouettes of the objects presented during the experiment. This mapping makes it possible to quantify the importance of the different regions on the visible surface of an object for its recognition by the participants.

Conclusions: The diagnostic attributes that have been identified are the best described in terms of surface fragments. Some of these fragments are located on or near the outer edge of the stimulus while others are relatively distant. The overlap is minimal between the effective attributes for the different points of view of the same object. This suggests that the traits underlying the recognition of objects are specific to the point of view. In other words, they do not generalize through the points of view.

AB055. Eye movements in the dark: saccades to non-visual targets

:-

Background: Saccades are rapid and abrupt eye movements that allow us to change the point of fixation very quickly. Saccades are generally made to visual points of interest, but we can also saccade to non-visual objects that attract our attention. While there is a plethora of studies investigating saccadic eye movements to visual targets, there is very little evidence of how eye movement planning occurs when individuals are performing eye movements to non-visual targets across different sensory modalities.

Methods: Fifteen adults with normal, or corrected to normal, vision made saccades to either visual, auditory, tactile or proprioceptive targets. In the auditory condition a speaker was positioned at one of eight locations along a circle surrounding a central fixation point. In the proprioceptive condition the participant’s finger was placed at one of the eight locations. In the tactile condition participants were touched on their right forearm in one of four eccentric location, left and right of a central point. Eye movements were made in complete darkness.

Results: We compared the precision and accuracy of the eye movements to tactile, proprioceptive, and auditory targets in the dark. Overall, both precision and accuracy of movements to non-visual targets were significantly lower compared to visual targets.

Conclusions: These differences emphasize the central role of the visual system in saccade planning.

AB025. Machine learning applied to the oxygen induced retinopathy model

:-

Background: The oxygen induced retinopathy rodent model is widely used, notably for the assessment of developmental dystrophies in preclinical studies of vascular retinal diseases. Typically, the quantification of vessel tufts and avascular regions is computed manually from flat mounted retinas imaged using fluorescent probes that highlight the vascular network. However, such manual measurements are time-consuming and hampered by user variability and bias, thus a rapid and objective alternative is required.

Methods: We employ a machine learning approach to segment and characterize vascular tufts. The proposed quantitative retinal vascular assessment (QuRVA) technique uses quadratic discrimination analysis and morphological techniques to provide reliable measurements of vascular density and pathological vascular tuft regions, devoid of user intervention within seconds. Our algorithms allow also delineating the whole vasculature network, and identifying and analyzing avascular regions.

Results: Our first experiment shows the high degree of error and variability of manual segmentations. In consequence, we developed a set of algorithms to perform this task automatically. We benchmark and validate the results of our analysis pipeline using the consensus of several manually curated segmentations using commonly used computer tools. We describe the method, provide details for reproducing the algorithm, and validate all aspects of the analysis.

Conclusions: Manual and semi-automated procedures for tuft detection present strong fluctuations among users, demonstrating the need for fast and unbiased tools in this highly active research field with tremendous implications for basic research and industry.

Structural analysis of processed corneas

:-

Background: Disruption of the microstructure in corneal stroma can lead to the loss of transparency. The lack of a characterization method for the microstructure prevents such scaffolds to be implemented in tissue transplantation. The non-invasive, three-dimensional (3D) rendering multiphoton microscopy (MPM) poses the potential to solve this problem.

Methods: MPM images and data analyses were performed with three kinds of samples with known and different quality. Isosurfaces (ISOs) were constructed for the evaluation of void volume and collagen distribution.

Results: The differences in the microstructures of these samples were revealed with clear indications and links to their behaviours in rehydration and possible transparency. According to this analysis, the scaffold with the highest void space ratio amongst the three presented the highest successful rates to be thoroughly rehydrated.

Conclusions: Such a method can be developed for assessing the quality of tissue engineered corneas, or donated corneas, and be useful as a powerful research tool in cornea related research.
