目的:探讨 miR-145-5p 调控视网膜 β-catenin 表达对小鼠眼轴的影响。方法:取 18 只雄性 周龄 C57BL/6 小鼠按照随机数字表法随机分成对照组,miR-145-5p mimics 组和 miR-145-5p 对照组。分组第 天双眼玻璃体腔注射 次药物,共注射 次 :对照组小鼠玻璃体腔内注射 3 μ枸橼酸盐缓冲液,miR-145-5p mimics 组小鼠玻璃体腔内注射 3 μ含有 3 μg miR-145-5p mimics 的枸橼酸盐缓冲液,miR-145-5p 对照组采取同样方法注射 3 μg miR-145-5p 对照载体。最后一次注射完成 3 d 后 超测量小鼠眼轴长度,之后取小鼠视网膜组织,采用 RT-PCR 技术检测视网膜中 miR-145-5p 和 β-catenin 的表达,采用 Western 印迹法检测视网膜组织中 β-catenin 的含量。取 只成年雄性 C57BL/6 小鼠,构建 β-catenin mRNA 3'UTR 荧光素酶过表达载体,将 β-catenin mRNA 3'UTR 荧光素酶过表达载体与 miR-145-5p mimics 共同转染小鼠视网膜组织,3 d 后取视网膜组织,应用荧光素酶报告基因技术检测和验证 miR-145-5p β-catenin 的相关性。结果:视网膜转染 miR-145-5p mimics 后,小鼠眼轴长度降低,与对照组相比有显著性差异P<0.05,视网膜组织中 miR-145-5p 表达上升,β-catenin 在 mRNA 和蛋白水平表达均下降P<0.05。视网膜转染 miR-145-5p 对照后,视网膜组织中 miR-145-5p 表达以及 β-catenin 的蛋白含量未见明显变化P>0.05;荧光素酶报告基因结果显示转染 miR-145-5p 和 β-catenin mRNA 3'UTR 荧光素酶过表达载体后,视网膜组织中荧光活性显著降低P<0.001)。结论:miR-145-5p 能够下调视网膜 β-catenin 的表达,进而控制眼轴增长。

Objective: To investigate the effect of miR-145-5p on mouse axial length by regulating the expression of β-catenin.Methods: Eighteen 3-week-old C57BL/6 mice were divided to the control, miR-145-5p mimics and miR-145-5p control groups according to the random number table. Both eyes received intravitreal injection once every 3 days, for 3 injections in total 2nd, 5th and 8th days. Patients in the control group received intravitreal injection of 3 μL citrate buffer, those in the miR-145-5p mimic group received intravitreal injection of 3 μL citrate buffer with 3 μg miR-145-5p mimics, and patients in the miR-145-5p control group received 3 μg miR-145-5p control. Axial length was measured by A-scan ultrasonography 3 days after the last intravitreal injection, then the expression of miR-145-5p and β-catenin in retinas was analyzed by RT-PCR or Western blot. After the over-expressed luciferase vector of β-catenin mRNA 3'UTR was constructed, the mRNA 3'UTR and miR-145-5p mimics were transfected to the retina of 6 male adult C57BL/6 mice. Luciferase reporter gene assay was used to verify the relationship between miR-145-5p and β-catenin in the retina. Results: After the transfection of miR-145-5p mimics in the retina, the expression of miR-145-5p in the retina was significantly up-regulated P<0.05, but the axial length and expression of β-catenin decreased significantly compared to those in the control group both P<0.05. There were no significant differences in the axial length, miR-145-5p and β-catenin expression after the transfection of miR-145-5p control. The results of luciferase reporter gene assay showed that the fluorescence activities in the retina were decreased significantly after the transfection of over-expressed luciferase vectors of β-catenin mRNA 3'UTR. Conclusion: miR-145-5p may regulate the axial length growth by inhibiting the Wnt/β-catenin pathway in the mouse retina.

中药超声雾化联合针灸治疗睑板腺功能障碍的临床疗效

目的:观察中药超声雾化联合针灸治疗睑板腺功能障碍(meibomain gland dysfunction,MGD)的临床疗效。方法:选取在新华医院就诊的7 7例(154眼)双 眼MGD患者,随机分为对照组(27例5 4眼)、中药超声雾化治疗组(雾化组;2 5例5 0眼)和中药超声雾化联合针灸治疗组(联合治疗组;2 5例5 0眼)。对照组接受睑板腺按摩治疗,雾化组在睑板腺按摩的基础上行中药超声雾化治疗,联合治疗组为进一步加用针灸治疗,治疗周期为4周。治疗前后收集患者双眼泪膜破裂时间(tear breakup time,BUT)、双眼基础泪液分泌试验数据(Schirmer I test,SIT)、双眼角膜荧光素染色评分(fluorescent,FL)、睑板腺分泌物性状评分(Meibomian Glandyielding Secretion Score,MGYSS)、睑缘异常评分(Eyelid Margin Abnormality Score,EMAS)以及主观症状积分等资料,并评价临床疗效。结果:治疗前三组BUT、SIT、F L、MGYSS、EMAS和主观评价得分等组间差异均无统计学意义(均P >0.05)。治疗4周后,各组受试者的BUT、SIT、F L评分、MGYSS评分、EMAS评分和主观评价评分与治疗前相比差异均具有统计学意义(均P < 0.05);联合治疗组的上述指标分别与雾化组及对照组相比,差异均有统计学意义(P <0.05)。治疗后对照组的总有效率为51.85%(28眼/54眼),雾化组为84%(42眼/50眼),联合治疗组为96%(48眼/50眼),联合治疗组总有效率与雾化组相比差异有统计学意义(P <0.05)。结论:中药超声雾化联合针灸治疗在改善MGD患者的睑板腺功能和减轻患者的干眼症状方面具有较好的临床疗效。
Objective: To observe the clinical efficacy of ultrasonic atomization fumigation of traditional Chinese medicine combined with acupuncture in the treatment of meibomian gland dysfunction (MGD). Methods: A total of 77 MGD patients (154 eyes) in Xinhua Hospital were selected and randomly divided into a control group (27 cases,54 eyes), a ultrasonic atomization of traditional Chinese medicine group (atomization group; 25 cases, 50 eyes),and a ultrasonic atomization of traditional Chinese medicine combined with acupuncture group (combined treatment group; 25 cases, 50 eyes). The control group received meibomian gland massage, the atomization group received meibomian gland massage and ultrasonic atomization of traditional Chinese medicine, and the combined treatment group received additional acupuncture treatment. The treatment period was 4 weeks. Tear breakup time (BUT), basal tear secretion test data (Schirmer I Test, SIT), corneal fluorescein staining score (FL), and Meibomian Gland Yielding Secretion Score (MGYSS), Eyelid Margin Abnormality Score (EMAS), Subjective Symptom Score (SSS) were collected and conduct clinical efficacy were evaluated. Results: There were no significant differences in BUT, SIT, FL, MGYSS, EMAS and SSS among the three groups (all P>0.05).After treatment of 4 weeks, the BUT, SIT, FL, MGYSS, EMAS and SSS in each group were improved significantly compared to baseline (P<0.05). The above indicators in combined treatment group were significantly different from atomization group (P<0.05) and control group (P<0.05). The total effective rates after treatment were 51.85% (28/54) in control group, 84% (42/50) in atomization group, and 96% (48/50) in combined treatment group. The difference in total effective rate between combined treatment group and atomization group was significant (P<0.05). Conclusion: Ultrasonic atomization of traditional Chinese medicine combined with acupuncture presents good clinical effects in improving the meibomian gland function of patients with MGD and alleviating the dry eye symptoms.
